Toward Universal Detection: Advancing Quantitative Analysis with Charged Aerosol Detection

In recent years, the use of quantitative liquid chromatography (LC) coupled with charged aerosol detection (CAD) has expanded rapidly, particularly for analyzing poor UV-absorbing analytes in multicomponent mixtures. As CAD continues to evolve toward becoming a near-universal detection method, recent research has focused on enhancing detector performance through response optimization. In this presentation, we highlight significant advancements in the linearity of the CAD detector achieved through the development of novel, in-house algorithm, which shows improved performance compared to existing approaches, including the default models implemented in commercial software. We further illustrate how this updated algorithm has been integrated into Amgen’s microgram high-throughput purification (µgHTPP) workflow, enabling robust quantitation of discovery-stage compounds. The presented model delivers more accurate real-world quantitation, positioning it for broader application in discovery settings, where authentic standards or response factors are unavailable.
