Novel Bioluminescent Tools for the Assessment of ADC Fc Function and Bystander Killing

Information

Antibody-drug conjugates (ADCs) are complex therapeutics with multiple mechanisms of action, including antigen-targeted cytotoxicity, Fc-mediated immune effects, and bystander killing. Comprehensive ADC characterization requires robust assays, but traditional methods often lack throughput, specificity, or ease of use. We introduce a suite of bioluminescent assays to streamline ADC assessment, including Fc receptor (FcR) binding, ADCC, ADCP, and bystander killing. Lumit® FcR Binding Assays offer a homogeneous, no-wash method for quantifying ADC-FcγRIIa, FcγRIIIa, and FcRn interactions. ADCC and ADCP Reporter Bioassays measure effector cell-mediated functions, while a HiBiT-based Bystander Killing Assay detects payload-induced lysis of antigen-negative cells. These assays differentiate ADCs like Enhertu and Kadcyla based on functional profiles. Our high-throughput, sensitive tools enhance ADC characterization, improving drug development and quality control with minimal handling and superior reproducibility.
