Information
InVivo provides custom services for the development of mouse hybridomas and the subsequent production of monoclonal antibodies against your antigen of interest.
Immunization (using proteins, peptides or haptens)
Starting material: 5 mg antigen of interest provided by customer (alternatively, the antigen can be produced by InVivo, in E. coli or in HEK/CHO cells via transient expression).
Immunization of three Balb/c mice, incl. boosting and bleeding
Detection of antibody titer in mouse serum by ELISA and selection of the highest-titer mouse for subsequent cell Fusion
Cell Fusion and Screening
Generation of hybridoma primary cultures by fusion of spleen cells from the selected mouse with the myeloma cell line SP2/0
Screening of primary cultures via ELISA for antibody Producers
Cell propagation and retesting of antibody titers to find clones
Limited Dilution Cloning, Screening and Characterization
Two-step-selection process to identify the two final clones (stable producers), via limited dilution cloning and antibody titer testing by ELISA
Cultivation and propagation of the two final clones
Isotyping and mycoplasma test
Cryoconservation
Deliverables:
Cryo-stored clones on dry ice (3 vials for each final clone)
Detailed protocol and documentation
Our standard procedure can be adjusted to individual customer needs.
Additional Services:
Welcome Offer: Cultivation in 1 L scale. One-Step Purification. Guaranteed yield of 10 mg purified monoclonal antibody.
Screening for antibody candidates for sandwich ELISA development
Negative screening to avoid cross-reactions of antibody with unspecific partners
