VyCAPVyCAP is a biotech company that developed the Puncher platform to screen single cells on the secretion of antibodies and proteins and isolate the single cell that produces the molecules of interest for clonal expansion, DNA and RNA isolation.    This Puncher platform allows screening thousands of single cells on their production of antibody within a couple of days, reduces the selection of the cells that produce the molecules of interest and cell line generation from months to weeks. The platform uses VyCAP proprietary microwell plate that contains 6400 individual microwells in an area of only 8 x 8 mm. Each of these wells contains a single cell that were pulled into these individual wells using microfluidic forces. Once the wells are filled with single cells, the secretion is measured by connecting the microwell chip with a capturing surface that is supplied with capturing ligands against the molecules of interest. The secreted molecules are made visible as dots using standard Fluorospot and ELISA methods. Based on their secreted molecules the microwell in which the cell resides is identified. Next, the cell of interest is isolated from its microwell by punching the bottom + cell from microwell chip towards your plate or tube of choice.  The Puncher platform:  · reduces the time for cell line generation from months to weeks  · allows screening of thousands of single cells on their production of antibodies in days  · assures 100% clonality  · is fully compatible with your current already developed protocols and reagents for Elispot and fluorospot single cell assays  · saves large amounts of culture media and plastics
